Calibration Curves

A column, which has the most appropriated calibration curve for the sample molecular weight range should be selected. For example, regarding the two columns (1) and (2), whose calibration curves are shown in the figure, column (2) is best suited for the analysis of the mixture of A, B and C. In other words, among the column calibration curves which covers all the sample molecular weight range, a column whose calibration curve has the smallest inclination should be selected to obtain a good separation.

THF Eluent Generally, polystyrene (PS) standards are used as calibration standards for organic SEC (GPC) with THF eluent.
However, when there is a large difference between the structure of the polymer sample and that of the polystyrene, other standards may be used, if the structure is close to that of the polymer. Poly(ethylene glycol) (PEG) or poly(methyl methacrylate) (PMMA) can be used for this purpose.
Chloroform Eluent PS standards are commonly used for organic SEC (GPC) analysis with chloroform eluent.
DMF Eluent Polyethylene oxide (PEO) and PEG are used for the calibration standards of organic SEC (GPC) analysis with DMF eluent, when using KD-800 columns. If PS is used for the standard, PS elutes slower because PS is adsorbed by the packing material of KD-800 columns.
Different from KD-800 columns, PS as well as PEG and PEO can be used as calibration standards for the SB-800 HQ series with DMF eluent. For samples, which cannot be analyzed by the KD-800 columns because of the adsorption by the packing material, the SB-800 HQ series can be employed.
Toluene Eluent PS is used for the standard for organic SEC (GPC) analysis with toluene eluent.
Quinoline Eluent PEG is used for the standard for organic SEC (GPC) analysis with guinoline eluent.
Methanol Eluent PEO and PEG is used for the standard for organic SEC (GPC) analysis with methanol eluent because they are easily dissolved in methanol.
Aqueous Eluent Pullulan, PEO and PEG are used for Aqueous SEC (GFC) analysis with aqueous eluents, such as pure water or aqueous salt solution.

GPC KF-806M and KF-806L are both packed with mixed type gel. However, compared with KF-806M, KF-806L which was developed recently has better linearity in the calibration curve.

Around the range of molecular weight 100, a solvent peak and/or other peaks coming from polymer additives may appear. For improving the separation in the low molecular weight range without affecting the separation in the high molecular weight range, it is recommended to use a solvent-peak separation column such as GPC KF-800DKF-600DKF-400D (for THF) or K-800D (for chloroform). The solvent-peak separation column should be used with linear type columns.

When an eluent with a relatively high salt concentration is used for Asahipak GS series, substances are eluted in decreasing order of molecular weight as it is the GFC mode which function mainly. GS-520, which has a large pore size, can analyze proteins with molecular weight up to 1,000,000. Samples such as human and bovine serum albumins which exhibit strong affinity with the molecular structure of the packing material are eluted belatedly. Making use of this characteristics, GS-520 is used exclusively for the analysis of serum albumin.

Several kinds of pydrophilic polymer standard are commercially available. When analyzing a sample, a standard which resembles it in structure should be selected to obtain the calibration curve for the analysis.

The calibration curves for downsized organic SEC (GPC) columns using PMMA are shown. In the case of the analysis for polymer whose MW is 1,000,000 or less, GPC LF-604 or LF-404 is suitable. When it is needed to separate low MW polymer and high MW polymer and low MW polymer is the purpose for the analysis, GPC HFIP-603 is suitable.
